ABSTRACT

OBJECTIVE: To observe the early adverse effect index caused by short-term-repeated exposure to cadmium chloride via oral perfusion in male rats. METHODS: Forty specific pathogen free healthy male SD rats were randomly divided into four groups: control group,low-,middle-and high-dose groups. The rats of low-,middle-and high-dose groups were treated with cadmium chloride 1. 11,3. 51 and 11. 06 mg/kg body weight, respectively, and the control group rats was treated with the same volume of ultra pure water,by gavage once a day for four weeks. During the experimental duration, the body weights of the rats were taken and activity status of the rats was observed. After the experiment, the rats were executed, and some indicators of main organ coefficients, blood routine, serum biochemical indexes, urine related effect indexes and bone mineral density were measured. RESULTS: During the experimental duration, rats of high-dose group showed the symptoms such as decreased activity, increase repose, move slowly and skin duller. Comparing with control group at the same time points, the body masses of the high-dose group of the 1-4 weeks were lower(P < 0. 05).After the experiment, comparing with control group, the weights of kidney and spleen of the high-dose group decreased significantly(P < 0. 05) and the liver coefficient increased significantly(P < 0. 05). The cadmium levels in blood, urine, liver, kidney and thighbone of the middle-and high-dose groups were higher than those of the control group(P < 0. 05).The red blood cell counts of the low-and middle-dose groups increased significantly(P < 0. 05). The level of hemoglobin of middle-and high-dose groups decreased(P < 0. 05),and the activity of alanine aminotransferase in high-dose groups increased significantly(P < 0. 05). Comparing with control group, the levels of urine α_1-microglobulin and urine β_2-microglobulin in urine of the middle-dose group were decreased(P < 0. 05) and the level of urine urea nitrogen increased(P < 0. 05),but there were no significantly changes of the above three indexes in the high-dose group(P >0. 05). There were no significant difference of the levels of N-acetyl-beta-D glucosaminidase in urine between control and treatment groups(P > 0. 05). Simultaneously,in high-dose group, the weight of thighbone, the bone calcium content and bone mineral density reduced significantly than those of the control group(P < 0. 05). CONCLUSION: Skeletal effects can be used as an early toxic effect sensitive index of short-term-repeated experiments exposure to cadmium chloride via oral perfusion in male rats.

ABSTRACT

Objective@#To investigate the determinants affecting the heart rate deceleration capacity (DC) in patients with dilated cardiomyopathy (DCM).@*Methods@#One hundred patients with DCM (DCM group) and 202 healthy subjects (control group) were respectively enrolled. Echocardiography and 24 hours electrocardiogram were performed in all subjects. DC value was compared between the two groups. Multiple regression analysis was made to evaluate the related determinants of DC ((age, sex, echocardiographic parameters including the left atrial diameter (LAD) and left ventricular ejection fraction (LVEF)).@*Results@#(1) DC value was significantly lower in DCM group than in control group( (4.40±2.03) ms vs. (7.30±1.81) ms, P<0.01), prevalence of DC value≤4.5 ms was significantly higher in DCM group than in control group (62% vs. 6%, P<0.01). (2) DC value in the DCM group decreased in proportion to increasing LAD dimension, DC value was (5.60±2.04) ms, (4.50±2.07) ms and (3.60±1.62) ms (P<0.05) in DCM patients with LAD≤40 mm, 40 mm<LAD≤50 mm and LAD>50 mm, respectively. (3) DC value in the DCM group was negatively related to the LAD (r=-0.366, P<0.01), positively related to the LVEF (r= 0.241, P<0.01), but not related with age and sex. Multiple factors regression analysis showed that increased LAD was related to the reduced DC values independtly.@*Conclusion@#DC value of the patients in the DCM group is decreased, which indicate the decrease of the vagus nerve tension, and increased LAD is related to the reduced DC value independtly in DCM patients.

ABSTRACT

Objective To investigate the most appropriate low radiation dose in multi-slice CT (MSCT) scans for orbital trauma patients. Methods Thirty trauma patients with suspected orbital fractures who underwent helical CT scans with a 64-MSCT using regular dose were selected. Noise was artificially introduced to the axial images using an image space noise addition tool to simulate 6 sets of lower dose scans with tube current of 30, 70, 100, 140, 170 and 200 mA, respectively. The lowest tube current with adequate image quality for confident diagnosis was determined based on the evaluation of the overall image quality and fracture detection on images at different dose levels. The determined lowest tube current was then validated using clinical scans. Radiation dose related parameters CTDIvol, DLP, ED were also recorded.Image quality was evaluated according to its low-density resolution, noise and structure clarity and characterized into 5-grades of excellent, good, fair, worse and worst Rank sum test and X2 test were wsed for statistics. Results In 30 trauma patients with regular dose of 300 mA, there were 30 cases of orbital fracture, 19 cases of intraorbital emphysema, 12 cases of ocular muscle injury and 1 case of intraorbital foreign body. These changes could still be clearly observed and correctly diagnosed when the tube current was reduced to as low as 70 mA. However, the overall image quality was mostly fair. At the simulated dose of 100 mA, the majority of images were characterized as excellent or good, and there was no statistical difference compared with that of regular dose scans (P > 0.05 ) . In the clinical evaluation for 20 orbital trauma patients with the reduced tube current of 100 mA, the majority of images were judged to be excellent (9 cases) or good(17 cases) and fair (4 cases). The radiation dose(0. 29 mSv) was reduced by 70% compared with that of regular tube current of 300 mA(0. 86 mSv). Conclusion The tube current of MSCT may be used as low as 100 mA in orbital trauma patients to reduce radiation dose and keep the image quality diagnosable.

ABSTRACT

Purpose To build up the Potassium Sodium Dehydroandroan Drographolide Succinate enteric coating sustained-release pellets delivery system by aqueous dispersion coating technique. Methods 1. Adopting MCC as vehicle, SiO_2 as antisticking agent, appropriate amount of 40% ethanol, preparing the core of pellets by extrude-spheronization method and the formulation and manufacturing process were optimized by orthogonal design. 2. Preparing the coating material with Eudragit L 30 D, which is used as pore-forming agent, EC as blocker and PEG6000 as plasticizer. The pellets were coated by fluid-bed coating method. Results 1. The optimal formulation and manufacturing process of pelltes' core were as follows: drug: MCC: SiO_2 = 7:7:5, extruding rate: 1 080 r/min, rounding rate: 960 r/min, rounding time: 5 min. 2. After the addition of EC: Eudragit L = 35:65, the plasticizer is 1.71 % and weight gain is 5% . The release in the gastric model fluid(pH 1.0) < 10% , and complete release ( > 80% ) in the enteric model fluid(pH 6.8) was in two hours. The release behavior accords with the regulations on the release rate of enteric preparation in ChP. Conclusion By adjusting the formulation and the parameters of the process of pellet and coating, we can make enteric coating Potassium Sodium Dehydroandroan Drographolide Succinate sustained-release pellets. All this accords with the regulation of pharmacopedia in vitro release.
